Abstract: Obtaining a good ¹H NMR spectrum requires an optimal sample amount. With a 400 MHz instrument and 5 mm tube, ~0.6 mL (~5 cm height) is recommended for maximum sensitivity and resolution. Much less volume lowers SNR and broadens peaks, but longer acquisition (more scans) can partially compensate.
